The Luminex ARIES M12 is an automated "sample-to-answer" molecular diagnostic system. To begin a run, start by scanning the barcode on the ARIES Assay Cassette. The system’s integrated barcode reader will automatically identify the test type (e.g., Flu A/B or C. diff) and the expiration date. It is critical to ensure that the sample has been properly liquefied and pipetted into the designated sample chamber of the cassette before it is inserted into the magazine.
Once the cassette is prepared, open the instrument door and slide the cassette into an empty slot in the M12 Magazine. The software will recognize the new hardware node and the "Slot Status" on the screen will change from Grey to Blue. You can load up to 12 cassettes for a single run, or start a "STAT" run for an individual sample if urgent results are required for the ER.
Finally, select "Start Run" on the xPONENT software interface. The system will perform a self-check of the Assay Modules to ensure the internal lasers and thermal cyclers are within operating parameters. Once the run begins, the door will remain locked until the process is complete (usually 90–120 minutes). Do not attempt to force the door open, as this will abort all active tests and waste the expensive chemical reagents.
